Investigation of the presence of sildenafil in herbal dietary supplements by validated HPLC method

INTRODUCTION: Sildenafil, a first FDA-approved PDE-5 inhibitor, is widely used in the treatment of erectile dysfunction due to its strong pharmacodynamic activity. Because many food supplements are nowadays incorporated into illegal adulteration processes, the identification of sildenafil in food supplements is crucial due to toxicological risks. A simple, fast, reliable high-performance liquid chromatographic (HPLC) method coupled with ultraviolet (UV) detector was developed and validated for the analysis of sildenafil in herbal dietary supplements (HDSs).
METHODS: The isocratic chromatographic separation was achieved by a C18 reverse-phase analytical column 4.6x250 mm, 5 µm particle size, using acetonitrile, 10 mM phosphate buffer containing 0.1% triethylamine (65: 35,v/v, pH 3.5) as a mobile phase. The mobile phase flow rate was 1 mL/min and the column temperature was 35°C. UV detector was set at 293 nm. A liquid-liquid extraction method further provided a simple and practical way to process HDSs containing sildenafil with a good recovery between 87.6-111.7%.
RESULTS: The method showed linearity with an excellent correlation coefficient above 0.999. Also, it was specific and sensitive by the limit of quantification value was 6.5 ng/mL. Intraday and inter-day method precision was below 8.2 (RSD%). Intraday and inter-day method accuracy were between (-4.0) to 7.1 (RE). The method was strong according to robustness test results obtained from UV detection, mobile phase buffer pH, column temperature and flow rate change values. This method was successfully applied to the 50 individual solid and liquid form HDSs.
DISCUSSION AND CONCLUSION: The results showed that the number of 37 out of 50 HDS sample (74%) contained sildenafil from 0.01 and 465.5 mg/g, 150.9 ±127.5 (mean ± SD), which could lead to serious health problems for its consumers might be fatal. The described procedure had been found to be simple, fast, precise and feasible for routine adulteration analysis of sildenafil, especially in food control or toxicology laboratories.

Bitkisel diyet takviyelerinde sildenafil varlığının valide bir HPLC yöntemiyle araştırılması

GİRİŞ ve AMAÇ: FDA tarafından onaylı ilk PDE-5 inhibitörü olan sildenafil, güçlü farmakodinamik etkinliği sebebiyle erektil fonksiyon bozukluğunun tedavisinde yaygın kullanılmaktadır. Günümüzde birçok gıda takviyesi, yasadışı tağşiş işlemlerine dahil olduğundan, gıda takviyelerinin içerisindeki sildenafil varlığının belirlenmesi toksikolojik riskleri sebebiyle çok önemlidir. Basit, hızlı, güvenilir bir ultraviyole (UV) detektörlü yüksek performanslı sıvı kromatografi (YPSK) yöntemi geliştirilmiş ve bitkisel gıda takviyelerinde (BGT) sildenafilin analizi için valide edilmiştir.
YÖNTEM ve GEREÇLER: . İçeriğinde % 0.1 trietilamin (65: 35, v / v, pH 3.5) bulunan asetonitril ve 10 mM fosfat tamponu mobil faz olarak, 5 µm partikül büyüklüğüne sahip 4.6 x 250 mm'lik bir ters faz C18 analitik kolonuna izokratik olarak uygulanmıştır. Mobil faz akış hızı 1 mL/dk ve kolon sıcaklığı 35°C idi. UV dedektör 293 nm'ye ayarlanmıştır. Çalışmada kullanılan sıvı-sıvı ekstraksiyon yöntemi, % 87.6 - 111.7’lik iyi geri kazanım değerleri ile BGT'lerin içerisinde bulunan sildenafilin yeniden elde edilmesi için kullanılan yöntem basit ve pratiklik sağlamıştır.
BULGULAR: Yöntemin doğrusallık testi sonuçlarından 0.999’dan büyük olmak üzere mükemmel bir korelasyon katsayısı (r2) elde edildi. 6.5 ng/mL'lik tayin limitiyle metod sildenafil belirlenmesi için seçici ve hassastı. Metodun gün içi ve günler arası hassasiyeti %8.2’nin (%RSD) altında idi. Metodun gün içi ve günler arası doğruluğu testi sonuçları (-4.0) ile 7.1 (RE) arasındaydı. Metodun UV saptama, mobil faz tampon pH'sı, kolon sıcaklığı ve akış hızı değişimlerinden elde edilen sonuçlarla sağlam olduğu görüldü. Bu yöntem, birbirinden farklı 50 adet katı ve sıvı formdaki BGT'ye başarıyla uygulandı.
TARTIŞMA ve SONUÇ: Sonuçlar, incelenen 50 BGT örneğinden 37'sinin (% 74), 0.01 ile 465.47 mg/g, 150.87±127.48 (ortalama±SD) arasında sildenafil içerdiğini ve gözlenen değerlerin kullanıcılarda ciddi sağlık sorunlarına yol açabileceğini gösterdi. Tanımlanan prosedürün, özellikle gıda kontrol veya toksikoloji laboratuvarlarında sildenafilin rutin tağşiş analizi için basit, hızlı, kesin ve uygulanabilir olduğu görülmüştür.
