The Effect of Ozone on Bone Strenght in Animal Model of Rheumatoid Arthritis

Introduction: Periarticular and systemic osteoporosis are more common in rheumatoid arthritis (RA) than normal population. In this study, we aimed to investigate the effectiveness of ozone on bone strength in Freund's Complete Adjuvant (FCA) arthritis, which is considered as the animal model for rheumatoid arthritis.
Material-method: In this study, 28 male Wistar rats were used. Saline was injected in hindpaws of half of these rats, and Freund’s complete adjuvant was injected in hindpaws of the other half of rats, subcutaneously. At the end of two weeks, 40µg/ml ozone (intraperitoneally) was administered for six times, within duration of 6 weeks in seven rats of each group. At the 6th week, serum IL-1, IL-6 and TNF-α levels were measured. Right femurs were separated for 3-D bending test.
Results: TNF-α levels of FCA arthritis was statistically higher than healthy group (p<0.05), but FCA arthritis- ozone group’s level was not (p>0.05). Serum levels of IL-1 and IL- 6 were not statistically significant amoung all groups (p>0.05). Maximum force and moment of inertia were upward trend in FCA arthritis-ozone group compare to the FCA arthritis group (p>0.05). The stifness and toughness were similar in the all groups (p>0.05).
Discussion: This is the first study in which the effects of ozone on bone strength of RA were investigated. It is determined that ozone is not effective enough, but not harmful on bone strenght of FCA arthritis. It is clear that further studies are required with ozone treatment and its use in RA when administrated in different doses and time courses.

Giriş: Romatoid artritte periartiküler ve sistemik osteoporoza normal populasyondan daha sık rastlanmaktadır. Bu çalışmada RA hayvan modeli olarak kabul edilen Freud’s complete adjuvan (FCA) artritinde ozonun kemik dayanıklılığı üzerine olan etkisini araştırmayı amaçladık.
Gereç-yöntem: Çalışmada 28 adet erkek Wistar rats kullanıldı. Bunların yarısının arka pençesine salin, diğer yarısına ise FCA subkutan enjekte edildi. 2 hafta sonunda her bir gruptan 7 rata, 3 hafta boyunca toplam 6 kez 40 μg/kg ozon (intraperitoneal) uygulandı. Sıçanların kan örneklerinde IL-1, IL-6 ve TNF-α düzeyleri değerlendirildi. 3 nokta bükme testi uygulanmak üzere sağ femurları çıkarıldı.
Bulgular: TNF-α düzeyi FCA artrit grubunda sağlıklı gruptan istatistiksel olarak yüksek bulunurken (p<0.05), FCA artrit-ozon grubunda bu değerler kontrol grubundan farklı bulunmadı (p>0.05). IL-1 ve IL-6 düzeylerinde ise gruplar arasında istatistiksel olarak anlamlı farklılık saptanmadı (p>0.05). Kemik dayanıklılığını değerlendirmede kullanılan paramatreler arasında maksimum kuvvet ve atalet momenti FCA artrit-ozon grubunda FCA artrit grubuna göre yükselme eğilimi göstermesine rağmen bu değişiklikler istatistiksel olarak anlamlı bulunmadı (p>0.05). Dayanım ve sertlikte ise gruplar arasında anlamlı farklılık saptanmamıştır (p>0.05).
Tartışma: Bu çalışma RA’da kemik dayanıklılığı üzerine ozonun etkisinin değerlendirildiği ilk çalışmadır. Ozonun RA hayvan modeli olan FCA artritinde kemik dayanıklılığı üzerine yeterince etkili olmadığı, ancak zarar da vermediği görülmüştür. Ancak ozonun farklı doz ve sürelerde uygulanması sonuçları değiştirebilir. Bu nedenle, bu konuda ilave çalışmalara ihtiyaç vardır.
